This book highlights super-resolution imaging using Stochastic Optical Reconstruction Microscopy (STORM). It begins with describing an optical microscope and its resolution limit due to the diffraction phenomenon. It also describes how diffraction barriers can be overcome by various optical super-resolution techniques. The book then discusses how super-resolution imaging can be achieved with the help of a simple experimental setup using the stochastic nature of fluorescence emission. It further elaborates the requirements in terms of excitation beam, fluorophore types, the imaging buffer and the sample preparation methods to observe fluorescence blinking, followed by a discussion on the image acquisition and the reconstruction algorithms. The book concludes with applications of STORM, its challenges and limitations.
