In the analysis for ion channel proteins, the demand for high screening throughput and long life-time has increased strongly. However, the available measurement systems still cannot meet the growing amount of the analytes for pharmaceutical industry or show an insufficient electrostability. In this work a microchamber based microelectrode cavity arrays (MECA) chip was developed to more toward a feasible solution for further increasing the throughput of ion channel screening with high resolution and long life-time. Black lipid membrane (BLM) formed on this chip was firstly electroporated to investigate the processes of membrane rupture and lipid pore formation which were related to the input electrical energy. Dielectrophoresis was applied to position the cell across the painted BLM based on this MECA chip. In order to perform whole-cell current recording based on this chip, trans-electroporation approach was applied to partially break the BLM and the cell membrane for a fusion between them. Ionic currents flowing through potassium rectifier (Kir) channels were successfully recorded with up to 1 GΩ sealing resistance of whole-cell configuration.
