Comparative histopathological study of bovine peripheral ganglia to identify factors affecting the risk of Bovine Spongiforme Encephalopathy
Aim of this study was the identification of factors that might modulate the risk of developing Bovine Spongiforme Encephalopathy (BSE) in dairy cattle from Lower Saxony. For that purpose, peripheral ganglia from a total of 441 cattle were examined histopathologically. Peripheral ganglia play an important role in the development of prion diseases. After oral infection, prions typically enter the body in in the gut, spread along nerve fibers and replicate in peripheral ganglia before entering the central nervous system.
These 441 cattle in this study include 22 animals with confirmed BSE, 13 suspected BSE-cases without detectable PrPSc-aggregates, 32 offspring and 270 animals from birth and feeding cohorts of BSE-cases that were culled for animal health reasons, as well as 104 cattle slaughtered for food. This study is unique in the number of included probable healthy dairy cows only slaughtered for animal health reasons. The selected ganglia include the celiac and cranial cervical ganglia as sympathetic ganglia and the trigeminal and nodose ganglia as cranial ganglia. In 41.1 % of all animals mononuclear inflammatory infiltrates were present in at least one ganglion with the celiac ganglion affected more often than the three other ganglia. No other types of inflammatory infiltrates, as suppurative or granulomatous, could be observed. Selected cases were tested for the presence of Herpesvirus, Bovine Astrovirus, Borna Disease Virus, Rabiesvirus, Listeria and double stranded RNA as a general marker for viral infections to identify common infectious causes of inflammatory infiltrates in nervous tissues. In none of the tested cases, an infection could be confirmed. A possible explanation for the presence of inflammatory infiltrates without detectable infectious pathogens might be a previous infection with inflammatory infiltrates as remnants of the immune system clearing the pathogens. Furthermore, non-infectious processes like metabolic diseases should be considered as origin of the described mononuclear infiltrates.
In addition, in 10.8 % of the celiac ganglia neoplasia could be observed. By histological and immunohistochemical features, these neoplasia could be typed as neurofibroma. Neurofibroma are benign peripheral nerve sheath tumors common in cattle and often found during meat inspection along the nerves of the brachial plexus, the thorax and mediastinum. In this study, bovine neurofibroma were described in the celiac ganglion for the first time.
Inflammatory infiltrates and neurofibroma could be found in cattle of all backgrounds. There was no evidence that BSE cattle are affected more often or severe by intraganglionic inflammatory infiltrates than other cattle in this study. Likewise, there was no evidence for a correlation of age and inflammation. Neither is for slaughtered cattle.
This study is the first to describe intraganglionic inflammatory infiltrates and neoplasia in dairy cattle from lower Saxony, independently from age and health status.
