This study consisted of 2 parts. In the first section, the plasmatic kinetics after oral loading on 6 beagles and the ideal dose of galactose were determined. 3 male neutered und 3 female neutered beagle with a median age of 47,3 months and median weight of 13,2 kg were examined. Afterwards, the kinetics of 6 dogs with portosystemic shunting were compared. These were 3 male and 3 female dogs, with a median age of 11,3 months and a median weight of 15,9 kg. The group consisted of following dog breeds: Retriever, Bernese mountain dog, Labrador, Shih Tzu and 2 crossbreeds. Blood samples of 6 fasted beagles were taken before and 15, 30, 45, 60 90 and 120 minutes after oral loading with 3 doses of galactose (0,25; 0,5; 0,75 mg/kg) combined with a repeated dose of 0,5 mg/kg. The 6 patients with portosystemic shunt underwent the same process with a dose of 0,5 mg/kg. Laboratory measurement was carried out with a published enzymatic/photometric method (galactose-dehydrogenase). Healthy dogs showed a good reproducibility of the dualistic kinetic and were comparable to humans. Basal there was no galactose in blood samples detectable. After oral loading a rise occurred with peak values after 30 to 45 minutes and decline until 120 minutes, without reaching initial values. Dogs with portosystemic shunts showed significant lower values as healthy dogs. There was a significant shift of the kinetics with later peak values. In summary, there is a significant difference in plasmatic kinetics of dogs with shunt compared to healthy dogs. Moreover, dogs with portosystemic shunt had lower values as expected. Further analysis should be made with attention to the renal elimination after oral loading with galactose.
